We have investigated the survival of PCR-amplifiable mitochondrial and nuclear DNA in a small number of modern and medieval bovine leather samples. The results of this preliminary investigation demonstrate that, while no nuclear DNA can be PCR-amplified from any of the specimens, mitochondrial DNA can be amplified from all samples. To investigate this contrasting pattern of DNA survival further, we have vegetable-tanned cow skin in our own laboratory, and directly assayed the survival of PCR-amplifiable mitochondrial and nuclear DNA at each step of the process. The results indicate that nuclear DNA is reduced to sub-amplifiable levels as a result of the tanning baths, whereas amplifiable mitochondrial DNA survives the complete process. Our results suggest that old and archaeological bovine leather may represent a useful source of genetic information, although this information will most likely be limited to that which can be gained from mitochondrial DNA.  相似文献

Ancient human migrations provide the critical genetic background to historical and contemporary human demographic patterns. However, our ability to infer past human migration events, especially those that occurred over rapid timescales, is often limited. A key example is the peopling of Polynesia, where the timing is relatively well defined, but the exact routes taken during the final stages and the source populations are not. Here, we discuss the technical limitations of current methods for inferring rapid human migration events, using the final stages of Polynesian migration as an example. We also introduce a promising new proxy method to infer human migrations—patterns of bacterial evolution within ancient dental calculus (calcified plaque). While we focus on Polynesia, this method should be applicable to other past migrations, enhancing our understanding of human prehistory and revealing the crucial events that shaped it.  相似文献

A human skull, buried about 2500 years ago in a Bronze Age cemetery at Jinggouzi, a site of an important ethnic group in ancient China, appeared to have characteristics of fibrous dysplasia. The CT images indicated a reduction in bone density and relatively homogeneous lesions. More features were revealed using CT reconstruction techniques. Lesions seen in low‐magnification images using a 3D deep‐field microscope had an irregular honeycomb‐like structure. At higher magnification, the trabeculae morphology and the gaps between the trabeculae were irregular and varied in size and shape. Paraffin‐embedded specimens stained with HE showed trabeculae with tortuous irregular arrangements varying in shape and width. The irregular trabeculae of woven bone has been described as having fibrous dysplasia. Molecular analysis of the GNAS gene indicated no mutation. This provides a non‐invasive approach for us to make more comprehensive diagnoses and to assist research into ancient human diseases.  相似文献

The study of ancient DNA (aDNA) using molecular methods is an increasingly valuable tactic in bioarchaeology. While this method must be carefully undertaken to ensure that the molecules detected are representative of the ancient sample and not modern contaminates, there is a danger that a ‘one size fits all’ approach to validation will lead to misinterpretation and/or missed opportunities of valuable findings. When comparing human and pathogen aDNA, there are many shared technical means that can ensure best practice. However, there are a number of assumptions that should not be used for both scenarios. We discuss these aspects in reference to a recent article published by this journal and highlight some of the latest advances in molecular detection of ancient pathogen DNA that can further improve this endeavour. Recovered and amplified ancient DNA (aDNA), from a historically documented 19th century Upper Canadian pioneer cemetery produced genotypes that were used to infer a past societal kinship system. While the results from multiplex short tandem repeat (STR) amplifications showed an unreliable polymerase chain reaction (PCR) product, a single locus HUMTH01 analysis yielded reproducible data and an allelic frequency pattern not statistically different from modern populations. Mitochondrial (mt) DNA HVR II data showed that a combined cemetery database exhibited reduced haplotype diversity indicators, as well as clusters of probable maternally related burials. The chronological persistence and replacement of mtDNA clusters approximately every two generations suggests a patrilineal/patrilocal kinship structure from a virilocal burial program for the Harmony Road cemetery. Through the integration of the aDNA analysis with archaeological material culture, historic records, and other ethnohistoric sources of information, this conclusion is supported. In this study persisting patrilineally inherited surnames act as a surrogate for aDNA Y‐chromosome haplotype analysis. These results suggest that aDNA applications on aggregate skeletal collections where sparse, or no ethnological or historical documentation exists, may result in incorrect population history inferences if the presence of a kinship interment bias is not considered. Recent zooarchaeological studies on water buffalo (Bubalus sp.) remains from China and south Asia question the traditional view that water buffalo were first domesticated in Neolithic China over 7000 years ago. The results from several recent population genetic studies of modern domesticated buffalo (Bubalus bubalis) are not consistent with each other, placing the original center of buffalo's domestication in south Asia, southeast Asia, or China. This paper reports a study using an ancient DNA approach to analyze water buffalo remains from Neolithic sites in north China to investigate their affinities with modern domesticated water buffalo, and to shed light on the origin of modern domesticated water buffalo in China.A 169 bp fragment of D-loop mitochondrial DNA was successfully amplified and verified for 13 of 24 bone samples obtained from seven archaeological sites along the Wei River valley in Shaanxi Province, China. The bone samples which yielded positive DNA can be dated to 8000–3600 cal. BP. The phylogenetic analysis of the obtained DNA sequences along with modern water buffalo sequences indicated that the ancient water buffalos were not the direct ancestor of modern domesticated water buffalo. However, the phylogenetic analysis, along with BLAST searches of these ancient DNA sequences, did demonstrate their relatedness to water buffalo more so than to any other bovid species, confirming the existence of indigenous wild (but now extinct) water buffalo species (B. mephistopheles) in ancient China.The DNA analysis of these ancient remains failed to establish direct links between modern domesticated water buffalo (B. bubalis) and indigenous water buffalo (B. mephistopheles) from ancient China. If further DNA studies of more ancient remains from other regions of China confirm the observation of solely indigenous water buffalo species in ancient China, it would suggest modern water buffalo might not have been first domesticated in China.  相似文献

薄荷醇作为新型的考古发掘现场临时固型提取材料,已经在秦俑一号坑的考古工地上得到了成功应用,但是关于薄荷醇实施提取的安全性能评估还需要深入研究。本实验开展了薄荷醇在秦俑一号坑考古发掘现场的遗迹(土壤和彩绘漆皮)上的实验室性能评估;采用压汞法研究了薄荷醇处理前后秦俑土颗粒的孔隙结构变化;采用超景深三维视频显微镜考察了薄荷醇施加并挥发过程中秦俑土和彩绘漆皮的显微结构变化;采用GC-MS分析了薄荷醇的残留;对薄荷醇挥发过程中在文物遗迹基底上的再结晶现象进行了讨论。研究结果表明,薄荷醇提取遗迹后不会破坏秦俑土的物理结构,彩绘漆皮形貌完好,作为考古发掘现场遗迹提取材料,薄荷醇在出土秦俑彩绘遗迹上使用时是安全的。  相似文献

It is believed on the basis of archaeozoological research that the domestic cat appeared in Central Europe during the Roman Period. In Poland, the domestic cat is a common species in medieval deposits. Only a few finds of cat remains of pre‐medieval age have been reported from Poland to date, including several specimens from deposits older than the Roman Period, dated to the pre‐Roman Period and even the Bronze Age. To clarify the earliest history of the domestic cat in Poland, the paper presents a review of the available published cat remains and adds some data about newly discovered remains. Combined methods of morphometry and ancient DNA were applied to enable distinction of wildcats and domestic cats. The domestic cat remains were radiocarbon dated. In six cases of domesticated cat reported in the literature, five were positively taxonomically verified, both by morphology and by genetic analysis, and one was recognised as a European wildcat. According to radiocarbon chronology, the oldest studied find is dated to the fourth–third century bc and represents a wildcat. Only two individuals of domestic cat – skeletons from Łojewo and Sławsko Wielkie, both from Kuyavia region (central‐northern Poland) – represent the Roman Period (first–third century ad ), and they are the oldest confirmed domestic cats in Poland.
