A principal problem facing human DNA studies that use old and degraded remains is contamination from other sources of human DNA. In this study we have attempted to contaminate deliberately bones and teeth sampled from a medieval collection excavated in Trondheim, Norway, in order to investigate this poorly understood phenomenon. Five pairs of teeth and bone samples were bathed in water containing various concentrations (from 10−9 and 10−21 g/l) of purified ΦX174 DNA. Subsequently the samples were subjected to a routine decontamination protocol involving a bleach bath followed by exposure to λ=254 nm ultraviolet light, prior to DNA extraction and analysis for evidence of the persistence of the contaminant. The results support previous speculation that bone is more susceptible to water‐borne sources of contaminant DNA, although both bone and teeth are readily contaminated and are difficult to decontaminate using the tested protocol. We believe that this is largely due to the porous nature of bone and teeth facilitating the deep penetration of the contaminant DNA. To simulate a more realistic handling situation, 27 further teeth were directly handled and washed, then decontaminated, prior to assaying for the residual presence of the handler's DNA. Surprisingly, although our results suggest that a large proportion of the teeth were contaminated with multiple sources of human DNA prior to our investigation, we were unable to contaminate the samples with further human DNA. One potential explanation may be the deposition of sediment or other structural changes that occur within the samples as they desiccate post‐excavation, which may protect samples from subsequent contamination, but also prevent the efficacy of bleach baths in decontaminating specimens. Much can be learned about the religious ideology and mortuary patterns as well as the demographic and health profiles of a population from archaeological human fetal skeletons. Fetal skeletons are rare, however, largely due to poor preservation and recovery, misidentification, or non‐inclusion in general burial populations. We present an analysis of 82 fetal/perinatal skeletons recovered from Kellis 2, a Roman Period cemetery dated to the third and fourth centuries AD, located in the Dakhleh Oasis, Egypt. Most of the fetal remains were individually wrapped in linen and all were buried among the general cemetery population in a supine, east–west orientation with the head facing to the west. Gestational age estimates are calculated from diaphysis lengths using published regression and Bayesian methods. The overall similarity between the fetal age distributions calculated from the regression and Bayesian methods suggests that the correlation between diaphysis length and gestational age is typically strong enough to avoid the ‘regression’ problem of having the age structure of reference samples adversely affecting the age distribution of target samples. The inherent bias of the regression methods, however, is primarily reflected in the gestational age categories between 36 and 42 weeks corresponding with the expected increase in growth variation during the late third trimester. The results suggest that the fetal age distribution at Kellis 2 does not differ from the natural expected mortality distribution. Therefore, practices such as infanticide can be ruled out as having a significant effect on the observed mortality distribution. Moreover, the Kellis 2 sample is well represented in each gestational age category, suggesting that all premature stillbirths and neonatal deaths received similar burial rites. The age distribution of the Kellis 2 fetal remains suggests that emerging Christian concepts, such as the ‘soul’ and the ‘afterlife’, were being applied to everyone including fetuses of all gestational ages. The reliable identification of fakes consisting of bronze often presents problems, because traditional methods such as stylistic studies, optical microscopy, chemical analysis or X‐ray diffraction of the corrosion may not be conclusive. We present a method that is based on the comparison of the tin isotope ratios 122Sn/116Sn and 117Sn/119Sn in the metal and in the adherent corrosion layer. An artificial patina is usually generated within a short time period. It has been observed that such a procedure leads to a depletion of the light tin isotopes in the corrosion layer, while in naturally corroded authentic archaeological objects no isotopic fractionation has been detected. The method has also been applied to archaeological objects and it could be confirmed, among other examples, that the famous ‘Sky Disc of Nebra’ is authentic. 相似文献
