The study focuses on the evidence for tuberculosis apparent in an Iron Age population recovered from the cemetery of Aymyrlyg, Tyva (Tuva), South Siberia. A recent wholly molecular study of five of the cases confirmed the presence of Mycobacterium tuberculosis (MTB) complex DNA in four of the individuals. In all cases the disease was caused by strains of Mycobacterium bovis rather than Mycobacterium tuberculosis and represents the first positive identification of the bovine form of the disease in archaeological human remains. Details of the palaeopathological characteristics of the cases are provided in the current paper, while the molecular observations are extended to include a quantitative evaluation of the surviving mycobacterial DNA using real-time PCR. The observation that bovine tuberculosis was the pathogen responsible is discussed in terms of current understanding of the evolution of the MTB complex as well as the implications for future ancient DNA studies in this area.  相似文献

Using ancient DNA methods, we have examined in detail two archaeological cases of leprosy from Mediaeval England. The first was a child skeleton with rhino-maxillary changes typical of lepromatous leprosy (LL). The second case was the skeleton of a male adult who showed both typical rhino-maxillary changes and osteitis/periostitis on the leg and foot bones. Bone powder was sampled from both cases and DNA extracts were prepared. These were subjected to a series of polymerase chain reactions (PCRs) specific for regions on the Mycobacterium leprae genome. The repetitive element RLEP was used for confirmation of M. leprae DNA and then three polymorphic regions were successfully amplified and sequenced to determine the number of variable nucleotide tandem repeats (vntr) at these loci. These were the microsatellite regions ML2344 and ML2172 and the minisatellite region ML0058. Genotyping data from the strains preserved within the skeletal remains were compared with those obtained for a reference strain of M. leprae. Variation at these three loci was found between both burials and the reference strain, indicating that vntr typing of LL cases from the archaeological record is a useful way of confirming disease and an additional means of authenticating aDNA data. This demonstrates the feasibility of targeting multiple loci for phylogenetic studies of leprosy strains from archival sources.  相似文献

The evolution of metallurgical practices over several millennia has resulted in human exposures to a diverse variety of toxic elements. These exposures were not just confined to the metalworkers, but in many cases broadly impacted the larger community and society. The role of lead in Roman civilization is particularly fascinating, where the available evidence of lead use in a myriad of applications by Roman civilization suggests the potential for elevated exposures. The present paper addresses the hypothesis that because of these practices, Roman occupants of ancient Londinium (43–410 ad ) were exposed to levels of lead that far exceeded exposures in the preceding populations of Iron Age Britain (700 bc –43 ad ). The elemental content and stable lead and strontium isotopic signatures of 30 femora from three Londinium cemeteries and radiocarbon-dated representative burials were examined and the results then compared with 70 femora dating to the pre-Roman Iron Age from the well-known Iron Age hillfort of Danebury, Hampshire, and from three cemeteries in East Yorkshire. Extensive efforts were directed at the minimization of sediment and soil contamination, the assessment of contamination from burial artefacts and a careful review of the burial context of specimens. Data for lead and 20 additional trace and major elements were obtained by magnetic sector inductively coupled plasma mass spectrometry (ICPMS), after acid dissolution of pre-cleaned bone cores. Lead isotope ratios in the bone core digests (and burial soils) were measured by multi-collector ICPMS. It was found that concentrations of lead in the Roman/Londinium-era femora were > 70-fold greater than those from pre-Roman populations. This was confirmed by femoral data obtained from the analysis of a pre-Roman burial from the early first century bc found adjacent to the Southern Cemetery. Several major and trace element metrics for diagenesis demonstrate that levels of lead in the bone are robust indicators of human exposure during life. Lead isotope data for the Roman population are consistent with previously identified widespread lead pollution. The pre-Roman populations contain lead isotopic compositions consistent with highly localized, minor sources of lead. Together, these data provide unequivocal confirmation that the Romans in Londinium were exposed to elevated lead levels. Elevated blood lead levels would have resulted, negatively affecting their health and possibly contributing to declining birth rates.  相似文献

The identification of sex in human remains recovered from archaeological locations is important in order to understand the social and biological structure of past societies, and to reconstruct past population demographic events. Sex determination is usually based on morphological traits of the skeletons, with the drawback that most methods do not apply to juveniles and require well preserved remains. In cases where morphological methods cannot be used, or are ambiguous, methods of molecular sexing systems are an alternative. In this methodological study we tested and validated the accuracy and usefulness of a molecular sexing method based on the amelogenin gene using pyrosequencing. We did this in a double blind study of documented 18th and 19th century human remains.  相似文献

Attempts were made to detect ancient DNA (aDNA) in samples of 88 human skeletons from eight Neolithic and Bronze Age sites in Greece and Crete. Ancient DNA was absent in specimens from Nea Nikomedia, Lerna, Karaviádena (Zakro), Antron Grave Circle A and Mycenae Grave Circle A. For each of three skeletons from Antron Grave Circle B that were sampled, polymerase chain reactions (PCRs) gave products for nuclear but not mitochondrial DNA, but amplicon yield was low and inconsistent with replicate PCRs failing to give reproducible results. With specimens from Mycenae Grave Circle B, evidence for mitochondrial aDNA was obtained for four of the 22 skeletons that were studied, and at Kouphovouno evidence for mitochondrial and/or nuclear aDNA was obtained with eight of the 20 skeletons that were examined. We conclude that, although aDNA might be present in some Eastern Mediterranean skeletons from later centuries of the Bronze Age, it is not commonly found in material from this period and is likely to be absent from older material.  相似文献

We report the recovery of short fragments of PCR amplifiable ancient DNA from exoskeletal fragments of the grain weevil Sitophilus granarius (L.) extracted from Roman and medieval deposits in Northern England. If DNA preservation in archaeological insect remains is widespread then many applications in the spheres of evolutionary studies and archaeology can be conceived, some of which are outlined.  相似文献

The presence of many phytolith-rich layers in late Bronze and Iron Age deposits at Tel Dor, Israel, are indicative of specific locations where plants were concentrated. Detailed studies of six of these phytolith-rich layers and associated sediments from Tel Dor show that the phytoliths were derived mainly from wild and domestic grasses. The most common domestic grass was the cereal Triticum aestivum (bread wheat). Three of these layers have a microlaminated microstructure, associated dung spherulites and phosphate nodules; characteristics that all point to the phytolith-rich layers having formed from dung in animal enclosures. In two of the layers, the microlaminated structure is absent while dung spherulites and phosphate nodules are present, suggesting that these too originate from dung that was not deposited in an enclosure. The sixth layer is microlaminated but does not contain spherulites. We thus cannot suggest a parsimonious explanation of its observed properties. Concentrations of burnt phytoliths are present in three locations, implying that dung was either burnt in situ or the ashes from burnt dung were redeposited. The transformation of dung accumulations into phytolith-rich layers involves a loss of organic material and hence a significant reduction in sediment volume, which is clearly apparent in the stratigraphy of some of the locations examined. The volume reduction can be observed in the macrostratigraphy and has important implications with regard to macrostratigraphic interpretation. The presence of abundant phytolith-rich layers on the tell has significant implications for the concept of ‘urbanism’ during these periods.  相似文献

It is sixteen years since the first detection of Mycobacterium tuberculosis DNA in archaeological specimens, yet the validity of findings continues to be questioned. Rigorous scientific scrutiny and debate is valuable and has led to a coalescence of procedures and precautions amongst those actively engaged in this work. It is disappointing that these good practices are not recognised by certain scientists whose primary expertise is in the related fields of archaeology, palaeopathology, and eukaryote ancient DNA. There is a danger that by constant repetition, disputable and inadequately justified concerns will assume the status of self-perpetuating myths and misunderstandings. We discuss these issues with reference to a recent article in this journal, in which clear peer-reviewed scientific data were specifically targeted as part of a general critique of the field of the palaeomicrobiology of tuberculosis. We believe we have given sufficient evidence and cogent argument to persuade the unbiased reader that the views in the critique by Wilbur et al. are unjustified.  相似文献

The recovery of ancient DNA from archaeological wheat samples under different preservation conditions was assessed using a number of genetic markers. It was possible to amplify nuclear DNA from desiccated grains but not from charred. The desiccated grain was from a pre-Hispanic grain silo in Gran Canaria and showed excellent DNA preservation, enabling the amplification of the ribosomal DNA markers IGS and ITS, the upstream region of the HMW-glutenin locus and single-locus nuclear microsatellites. Our results demonstrated the presence of both durum and bread wheat in an assemblage of naked grain. We were also able to identify different genotypes in durum wheat and compare these with extant landraces, providing insights into the agrarian practices of the ancient Canarians and the origin of their crops.  相似文献

Climate deterioration at around the time of the Bronze Age/Iron Age transition has for long been argued to have resulted in upland abandonment in northern and western Britain, and recent research has provided evidence that a major climate downturn from 850 cal BC caused settlement abandonment in western Europe and potentially worldwide. It is, however, unclear to what extent only ‘marginal’ sites were affected, due to the lack of any systematic attempt to view the evidence for settlement and land-use change across a range of landscape types with differing sensitivities to environmental change. This paper addresses this issue by an evaluation of 75 pollen sequences spanning the late Bronze Age and early Iron Age in Britain to assess whether climatic deterioration was sufficient to cause widespread land abandonment. The results provide no evidence for wholesale land-use change at this time; the overall picture is one of continuity of land use or even increased agricultural activity. There are, however, hints of regional variability, with a greater tendency to abandonment of upland areas in Wales, and signs of woodland regeneration in agriculturally productive areas of lowland central southern England. The latter pattern may reflect a combination of rising ground-water levels affecting local land-use in the immediate vicinity of the mires which provide the source of the pollen data, against a backdrop of regional-scale social and economic changes at the Bronze Age-Iron Age transition.  相似文献

Stratified societies in southern Africa first developed in the Shashe–Limpopo Basin. As is well known, rank-based society at K2 developed into class distinction at Mapungubwe. The transfer of this new social organisation to Great Zimbabwe has received less attention. New research on rainmaking practices suggests that a Mapungubwe dynasty introduced class structures at Great Zimbabwe. Poor climatic conditions at the end of the 13th century undermined sacred leaders at Mapungubwe itself, and while vulnerable, the elite at Great Zimbabwe took over the important gold and ivory trade. Among other things, the new elite used the unique Zimbabwe birdstones to establish their legitimacy.  相似文献

Occipital bone lesions on an Iron Age horse cranium from the burial mound of Arzhan 1, Tuva, Central Asia, are described and interpreted. Cavitations around the nuchal ligament attachment site on the skull are interpreted as foci of inflammation and necrosis following local infection. It is suggested that the pathology represents a case of ‘poll‐evil’, most likely due to a bacterial infection. The significance of such an interpretation is discussed, including its implications for disease ecology and the possible infection risks to contiguous animal and human communities of the first millennium BC in Central Asia. An osteometric scoring technique is developed for the quantification of rickets deformational effects persisting in the healed adult skeleton. Termed RR-15 scoring (for residual rickets estimate, based on 15 main diagnostic osteometric traits), the method is first validated for sunshine deficit, and is then tested in a comparison of three climatically distinct archaeological populations from southeastern England. Two well-dated cemetery series were sampled: the Saint Bride's Church columbarium collection (SBC) from London at the time of the Industrial Revolution, for which the sex, age, and calender year at death of each individual are known; and the early Anglo-Saxons from Abingdon, near Oxford (AAS). Estimates of insolation in the past were developed indirectly by reference to δ18O mass spectrometer analyses of dated layers of the Greenland ice sheet. In SBC and AAS, the RR-15 score varies primarily as a direct function of computed sunshine deficit, but is also incremented by deficits of bioavailable calcium in their reconstructed diets, and by demographic stress in AAS, the inferred result of a high birth rate. The amounts of interglobular dentine present in the permanent first and third molar crowns of SBC and AAS have been shown to correlate similarly with sunshine and calcium deficits. The relatively high RR-15 score obtained in a small sample of northwestern European neanderthals lends quantitative support to the bioenvironmental hypothesis that sunshine-deficit rickets accounts for much of their paranormal gross morphology.  相似文献

The small mineral-binding bone protein, osteocalcin, has been applied in a number of studies on ancient bone due to predictions of its long-term stability. However, the intact protein has not been shown to survive in ancient bone devoid of DNA, which is a much more phylogenetically informative biomolecule. In this investigation, the survival of osteocalcin is directly compared to the amplification of mtDNA in a set of 34 archaeological samples from four sites throughout Europe. We also present unpublished osteocalcin sequences of seven mammalian species in addition to the 19 published sequences to highlight phylogenetic limitations of this protein. The results indicate that the intact osteocalcin molecule survives less in archaeological samples than mtDNA and is more subject to the temperature of the archaeological site. Amino acid analyses show the persistence of the dominant protein collagen in samples that failed both osteocalcin and mtDNA analyses. The implications these findings present for biomolecular species identification in archaeological and palaeontological material are that, although proteins do survive beyond ancient DNA, osteocalcin does not appear to be the most ideal target.  相似文献

Abstract

Middens of the southern British late Bronze and Iron Age are vast accumulations of cultural debris that can be explained as refuse dumps linked with large periodic feasting events. A distinctive feature of these sites is that their faunal assemblages invariably comprise a considerably higher proportion of pig remains than contemporaneous settlement sites. This paper presents results from a programme of stable carbon (δ13C) and nitrogen (δ15N) isotope analysis of fauna from two major midden sites, Llanmaes in South Wales and Potterne in Wiltshire. The research aim is to reconstruct husbandry strategies and foddering regimes, particularly concerning pigs, to better understand how the challenges of raising large herds were met. Analysis produced exceptionally wide-ranging results for pigs and other domesticates at both sites, particularly in terms of δ15N values, demonstrating that diverse foddering strategies were employed. Diversity in the late Bronze Age pig foddering regimes indicates that the Neolithic husbandry practices (focusing on woodland fodder) had not been abandoned, but that new husbandry methods (consumption of household waste) were also being practised, which subsequently became more widely established in the Iron Age. The heterogeneity of signatures suggests that animals may have been husbanded in a piecemeal fashion at a local, household level. This in turn hints that fauna may have been brought to these sites from households across the surrounding landscape, rather than being husbanded by specialist producers in the vicinity of the middens.  相似文献

Strontium isotope analysis represents an effective means of assessing mobility and reconstructing geographic residence patterns in archaeological populations. This biogeochemical technique was utilized to test the hypothesis that burgeoning interregional exchange networks and the occurrence of exotic grave goods in local tombs would correspond with a highly mobile population and a considerable immigrant presence during the Umm an-Nar (2700–2000 BC) period in the UAE. This region has been considered peripheral relative to larger civilizations in Mesopotamia and the Indus Valley, but played an important role as a major supplier of copper for the Persian Gulf. Individuals (n = 100) from six monumental Umm an-Nar tombs (Mowaihat; Tell Abraq; Umm an-Nar Island I, II, V; Unar 1) were selected to evaluate the geographic origins of tomb members. Mean 87Sr/86Sr ratios from local individuals interred at Mowaihat (0.708863 ± 0.000014; 1σ, n = 12), Tell Abraq (0.708873 ± 0.000020; 1σn = 27), Umm an-Nar Island (0.708902 ± 0.000079; 1σ, n = 33), and Unar 1 (0.708805 ± 0.000065; 1σ, n = 25) all display little isotopic variability, indicative of a population that was not highly mobile. However, coupled with archaeological evidence, three immigrants from Tell Abraq (n = 2) and Mowaihat (n = 1) identified by deviant strontium values suggest that this region was actively engaged in interregional interaction. Despite claims that these tombs acted as visible markers of territoriality legitimized by local ancestors buried within them, the presence of non-locals suggests that as commerce became increasingly important, definitions of kinship and social identity may have become more flexible to better meet the needs of the local community and those with whom they interacted.  相似文献

A metallurgically-oriented excavation in Area A at Tell es-Safi/Gath yielded evidence for iron and bronze production dating to the early Iron Age IIA. Two pit-like features, which differed considerably from one another in colour, texture and content, were excavated. Evidence shows that each feature represents a different in situ activity related to iron production, inferred by the presence of hammerscales, slag prills and slag. An upturned crucible was found on top of one of the features. Analysis of the crucible slag showed that it was used for bronze metallurgy. Tuyères, both round and square in cross-section, were found in and around the two features. The presence of the two industries together presents a unique opportunity to explore the relationship between copper and iron working. This is especially important against the background of the scarcity of evidence for iron production in the Levant during the early phases of the Iron Age.  相似文献

Launch of Very High Resolution (VHR) satellite sensors meant a significant evolution for Remote Sensing archaeological applications since data with high spatial resolution may be used to identify signals related to buried archaeological remains (e.g. cropmarks, soilmarks, dampmarks) in very extended territories. The aim of this work is to develop a suitable image processing workflow to improve the cropmark visibility in the VHR satellite images. The workflow implemented consists of two steps: first the panchromatic and multispectral images have been pansharpened in order to merge the two images, then some Enhancement methods have been applied on the pansharpened image. As a result of the several tested pansharpening techniques, the Principal Component Resolution Merge yields images of very good quality useful not only for visualization but also as input for further processing. Among the different enhancement methods taken into consideration, the Spatial Enhancement, carried out by applying the Wallis Adaptive filter further improve the visibility and recognition of the archaeological signals.  相似文献

Using multivariate techniques, several skulls of fossil large canids from sites in Belgium, Ukraine and Russia were examined to look for possible evidence of the presence of Palaeolithic dogs. Reference groups constituted of prehistoric dogs, and recent wolves and dogs. The fossil large canid from Goyet (Belgium), dated at c. 31,700 BP is clearly different from the recent wolves, resembling most closely the prehistoric dogs. Thus it is identified as a Palaeolithic dog, suggesting that dog domestication had already started during the Aurignacian. The Epigravettian Mezin 5490 (Ukraine) and Mezhirich (Ukraine) skulls are also identified as being Palaeolithic dogs. Selected Belgian specimens were analyzed for mtDNA and stable isotopes. All fossil samples yielded unique DNA sequences, indicating that the ancient Belgian large canids carried a substantial amount of genetic diversity. Furthermore, there is little evidence for phylogeographic structure in the Pleistocene large canids, as they do not form a homogenous genetic group. Although considerable variation occurs in the fossil canid isotope signatures between sites, the Belgian fossil large canids preyed in general on horse and large bovids.  相似文献

New field and laboratory work has been carried out on two vitrified forts, Rhubh Aird Ghamhsgail near Arisaig and The Torr near Acharacle. Fieldwork confirms that both are constructed from rubble largely comprising local psammitic Moine Supergroup rocks and that their walls are relatively well preserved with large portions of vitrified material remain, particularly at The Torr, where remnants of up to four courses of stone facing occur. The preserved wall fills comprise mostly psammitic rubble set in a dark brown, vesicular, glassy to aphanitic matrix that in many places preserves evidence of downward flow. Conventionally, vitrification is explained through combustion of timber interlacing within the walls leading to melting that has been modelled in the granite system. However, considering Iron Age smelting technology, this leads to problems in obtaining high temperatures (≥1000 °C) over tens of metres of wall length. Geochemical analysis of the vitrification indicates that modelling the melting within the granite system is incapable of providing a partial melt matching the composition of vitrification. At The Torr, pelite fragments have been found near the base of the wall where partial melt occurs as black bubbles. In these fragments it appears that micas (largely biotite) have formed the melt via the reaction biotite + quartz = sanidine + orthopyroxene + liquid which takes place at temperatures c. 850 °C. Quench crystals in the melt include orthopyroxene, plagioclase, spinels, ilmenite, magnetite, anatase and apatite confirming that it was not a granite melt. These melted fragments are found next to material with biotite that, superficially, appears unmelted, suggesting that some of the heating took place perhaps away from the edifice and unmelted residue was utilised as part of the rubble fill, suggesting that in situ partial melting of the rubble is unlikely. However, in other parts of the edifice heat affected, yet unmelted, fragments are dominant. The new evidence indicates that vitrification occurred at lower temperatures than previously modelled and thus the melts could have been achieved more easily than previously thought.  相似文献
