Domestic horses played a pivotal role in ancient China, but their exact origin remains controversial. To investigate the origin of Chinese domestic horses, we analyzed mitochondrial DNA (mtDNA) from 35 horse remains, aged between 4000 and 2000 years, excavated from nine archaeological sites in northern China. The Chinese ancient horses exhibited high matrilineal diversity, falling into all the seven haplogroups (A–G) observed in modern horses. These results suggest that several maternal lines were introduced into the gene pool of Chinese horses in the past. Haplogroups A and F were more prevalent in ancient horses than the other haplogroups. Interestingly, only haplogroups A and F were present in the samples older than 4000 years, while the more recent horses (between 2000 and 3000 years BP) fell into all seven haplogroups. Comparison with DNA data of present-day horses suggests that haplogroup F is like to be an ancient haplogroup of East Asian origin. These analyses also suggest that the origin of Chinese domestic horses is complex, and external mtDNA input occurred after initial domestication. Our results indicate that the Chinese ancient horses are more related to the modern Mongolian horses. Lastly, our results cannot support the previous hypothesis that early Chinese domestic horses were derived from the Przewalski horse.  相似文献

The search for the origins of syphilis has a long history in the medical and anthropological literatures. If we know more about the emergence of the pathogen that causes the disease in humans we will understand its evolution through time and space as well as shed light on its current state in living populations. Ancient DNA techniques used to isolate Treponema pallidum subsp. pallidum DNA from archaeological human specimens provide direct evidence of its existence in the past. However to date, only Kolman et al. (1999) have been successful in this endeavour, while other attempts have failed (e.g., Barnes and Thomas, 2006; Bouwman and Brown, 2005). Why has there been little success? This paper serves to compliment and add relevant information to Bouwman and Brown's and Barnes and Thomas' discussion concerning our inability to apply ancient DNA techniques to study venereal syphilis in past human populations.Our approach utilized 15 different human specimens from different geographies and different temporal periods: eight samples come from medically diagnosed individuals archived during the American Civil War period; six originate from the United Kingdom and predate 1492 with four of these samples having been previously analyzed by Bouwman and Brown and one sample comes from historic Canada. Human mitochondrial and amelogenin DNA, as well as several genes from the Treponema organism were analyzed revealing the relatively good preservation of human multi-copy and single copy DNA but not treponemal DNA. This study also incorporates a unique molecular experiment using rabbits infected with venereal syphilis to help illustrate that treponemal DNA disseminates to bone early during the first stages of infection but is not present in later stages of the disease using the techniques presented in this study.  相似文献

‘Historical’ DNA obtained from specimens preserved in natural history collections has proven useful for addressing a wide variety of questions, such as the spread of domesticated species or changes in genetic diversity. With the development of high‐throughput sequencing techniques, there is an increasing focus on acquiring genetic information encoded by single‐copy nuclear DNA from historical DNA extracts. The development of efficient techniques to determine the level of nuclear DNA preservation in candidate specimens is necessary to maximize the data obtained from these analyses. Although current evidence suggests that a sample's mitochondrial DNA preservation predicts its nuclear DNA preservation, we show that the relationships between mitochondrial and nuclear DNA preservation are complex.  相似文献

The use of wool derived from sheep (Ovis aries) hair shafts is widespread in ancient and historic textiles. Given that hair can represent a valuable source of ancient DNA, wool may represent a valuable genetic archive for studies on the domestication of the sheep. However, both the quality and content of DNA in hair shafts are known to vary, and it is possible that common treatments of wool such as dyeing may negatively impact the DNA. Using quantitative real-time polymerase chain reaction (PCR), we demonstrate that in general, short fragments of both mitochondrial and single-copy nuclear DNA can be PCR-amplified from wool derived from a variety of breeds, regardless of the body location or natural pigmentation. Furthermore, although DNA can be PCR-amplified from wool dyed with one of four common plant dyes (tansy, woad, madder, weld), the use of mordants such as alum or iron leads to considerable DNA degradation. Lastly, we demonstrate that mtDNA at least can be PCR-amplified, cloned and sequenced from a range of archaeological and historic Danish, Flemmish and Greenlandic wool textile samples. In summary, our data suggest that wool offers a promising source for future ancient mitochondrial DNA studies.  相似文献

Excavations at Cladh Hallan, a Bronze Age-Iron Age settlement on South Uist in the Outer Hebrides off the west coast of Scotland, revealed the skeletons of two adults, a sub-adult and a child buried beneath the foundations of three roundhouses. Osteological and isotopic evidence has shown that the male adult skeleton is a composite made up of parts of at least three different individuals. To test the hypothesis that the female skeleton was also a composite we examined ancient DNA from four of its components: the skull, mandible, right humerus and right femur. Seven polymerase chain reactions (PCRs) were attempted, these covering positions 15,996–16,420, 16–132 and 232–368 of the mitochondrial DNA hypervariable I and II regions. Three PCRs were successful for each sample and a total of 55 sequences were obtained from the cloned products. After exclusion of possible contaminating sequences, the remaining 34 were compared. It was concluded that the mandible, humerus and femur come from different individuals. Insufficient data were obtained to draw conclusions regarding the origin of the skull. The presence of two composite skeletons at Cladh Hallan indicates that the merging of identities may have been a deliberate act, perhaps designed to amalgamate different ancestries into a single lineage.  相似文献

Using ancient DNA methods, we have examined in detail two archaeological cases of leprosy from Mediaeval England. The first was a child skeleton with rhino-maxillary changes typical of lepromatous leprosy (LL). The second case was the skeleton of a male adult who showed both typical rhino-maxillary changes and osteitis/periostitis on the leg and foot bones. Bone powder was sampled from both cases and DNA extracts were prepared. These were subjected to a series of polymerase chain reactions (PCRs) specific for regions on the Mycobacterium leprae genome. The repetitive element RLEP was used for confirmation of M. leprae DNA and then three polymorphic regions were successfully amplified and sequenced to determine the number of variable nucleotide tandem repeats (vntr) at these loci. These were the microsatellite regions ML2344 and ML2172 and the minisatellite region ML0058. Genotyping data from the strains preserved within the skeletal remains were compared with those obtained for a reference strain of M. leprae. Variation at these three loci was found between both burials and the reference strain, indicating that vntr typing of LL cases from the archaeological record is a useful way of confirming disease and an additional means of authenticating aDNA data. This demonstrates the feasibility of targeting multiple loci for phylogenetic studies of leprosy strains from archival sources.  相似文献

We have investigated the survival of PCR-amplifiable mitochondrial and nuclear DNA in a small number of modern and medieval bovine leather samples. The results of this preliminary investigation demonstrate that, while no nuclear DNA can be PCR-amplified from any of the specimens, mitochondrial DNA can be amplified from all samples. To investigate this contrasting pattern of DNA survival further, we have vegetable-tanned cow skin in our own laboratory, and directly assayed the survival of PCR-amplifiable mitochondrial and nuclear DNA at each step of the process. The results indicate that nuclear DNA is reduced to sub-amplifiable levels as a result of the tanning baths, whereas amplifiable mitochondrial DNA survives the complete process. Our results suggest that old and archaeological bovine leather may represent a useful source of genetic information, although this information will most likely be limited to that which can be gained from mitochondrial DNA.  相似文献

The present case displays severe antemortem skeletal malformations suggestive of antemortem reactive arthritis (ReA), spondyloarthropathies and diffuse idiopathic skeletal hyperostosis (DISH). The ossified anterior longitudinal ligament on the right side of the thoracic vertebral bodies, with the presence of extraspinal enthesophytes, together indicates DISH. Fused lumbar vertebrae with costovertebral and sacroiliac joint fusion, combined with numerous large enthesophytes collectively suggest ankylosing spondylosis and ReA. Finally, some vertebrae showed evidence of age related osteoarthritis. However, the anatomical evidence for these conditions displays some inconsistencies with the known pathogenesis; therefore, a definitive account of this individual's antemortem condition remains uncertain. As no biological or medical information is known about the individual, skeletal morphology, irregular skeletal fusion and the presence of enthesophytes have been collectively used to propose the antemortem disorders that appear to have affected this individual's skeleton. Skeletal weight and/or weight of the different bones of the human skeleton are currently used in a wide range of applications such as archaeological cremations and forensics. Still, few reference values are available that compare the mean weights for the different skeletal parts. In this paper we present new reference values for total skeletal weight, including the weight of the different skeletal bones based on a modern Portuguese Identified Skeletal Collection (CEI) curated in the Museum of Anthropology of the University of Coimbra (Portugal). The mean weight of the entire skeleton for the CEI pooled sample is 3323.8 ± 779.6 g. Sex and age differences were investigated. As expected, males display heavier bones, at a statistically significant level. The mean weight of the male skeletons is 3850 g, and 2797.6 g for the female sample. Age differences were found, especially for the female samples in the 29–39 versus 50–59 and/or >60 age groups, probably as a consequence of age‐related bone loss in post‐menopausal women. For the male sample, no clear age‐related trend was observed but for the unexpected result that the second highest bone weight recorded is in the oldest age group (>60 years). This could result from high mechanical loadings and thus greater bone robusticy and amount of cortical bone. The percentage of weight of the different parts of the skeleton was also calculated. These new values can be quite useful in the study of very fragmentary and/or commingled human remains, recovered from historic or forensic contexts, since the complete identification, by side, of the remains is not necessary. Ancient DNA (aDNA) analysis was employed to determine the sex of slave sacrifice victims from Qin State tombs during the Spring and Autumn Period of China. It is difficult to obtain sex information from fragmentary skeleton samples with the aid of skeletal morphology methods. aDNA was extracted from the dentine in selected tooth samples of sacrificial slaves using a modified traditional method, which combines together phenol/chloroform extraction, silicon dioxide adsorption and ultrafiltration concentration. Based on the sequence differences between the amelogenin homologous gene in the X and that in the Y chromosome, a pair of specific primers was designed to identify the sex of the selected samples. In the selected eight typical samples, the aDNA analytical results revealed that three were males and two were females. These findings indicate that molecular sex identification might provide more valuable information for archaeological research on the institution of slave sacrifice in the Spring and Autumn Period of China.  相似文献

William Charles Macready's farce The Irishman in London; Or; The Happy African (1792) can be read as an exploration of Englishness in its relationship to Irishness as presented on the London stage in the period following the French Revolution. This article examines Macready's play as a critique of the common identification of Irishness with blackness that uses stock characters and attitudes to examine English identity. Through a marriage plot displaced from the traditional romantic heroes onto their Irish and black servants, we see English as a commercial identity rather than a cultural one. Linking race to culture and culture to nationality, Macready's play presents Irishness and blackness as culturally rich while calling into question the content of Englishness.  相似文献

This study evaluates a method for obtaining stature estimates for populations represented by skeletal material, with individuals buried in a supine position. During the excavation of a Danish mediaeval cemetery, in situ skeletal length in the grave was measured from a point above the cranial point farthest from the body to the most distal point of the talus. The measurement was made with a folding rule placed on the sagittal midline of the skeleton, allowed to follow any curvature of the skeleton in situ. In the laboratory, stature was reconstructed anatomically, and this stature was regarded as an accurate estimate of living stature. Stature was also reconstructed from femur length by two linear regression procedures: 1) by sample and sex specific formulae, employing a leave‐one‐out approach, and 2) by sex wise formulae for Euro‐Americans from Trotter & Gleser (1952, American Journal of Physical Anthropology 10 : 463–514). Skeletal length in the grave and the two stature estimates based on linear regression were compared to anatomically reconstructed stature. Skeletal length in the grave estimated anatomically reconstructed stature with practically no bias (95% CI: −1.3–1.5 cm). Sample specific regression formulae estimated anatomically reconstructed stature also with no bias (95% CI: −1.2–1.1 cm). In contrast, statures calculated from Trotter & Gleser's regression formulae estimated anatomically reconstructed stature with a bias of about 4 cm (95% CI: 3.3–5.0 cm). Estimates of stature variance were biased for all three estimation procedures. However, for samples of adults, an adjusted variance estimate can be obtained by subtracting 8.7 cm2 from the variance obtained from skeletal lengths in the grave. It is recommended to measure skeletal length in the grave whenever possible, and use this measurement for estimating statures for prehistoric and early historic populations. Immigrant integration is currently a prominent issue in virtually all contemporary democracies, but countries in which the historic population itself is deeply divided – particularly those with substate nations and multiple political identities – present some interesting questions where integration is concerned. The existence of multiple and potentially competing political identities may complicate the integration process, particularly if the central government and the substate nation promote different conceptions of citizenship and different nation‐building projects. What, then, are the implications of minority nationalism for immigrant integration? Are the added complexities a barrier to integration? Or do overlapping identities generate more points of contact between immigrants and their new home? This article addresses this question by probing immigrant and non‐immigrant ‘sense of belonging’ in Canada, both inside and outside Quebec. Data come from Statistics Canada's Ethnic Diversity Study. Our results suggest that competing nation‐building projects make the integration of newcomers more, rather than less, challenging.  相似文献

Existing methods to extract, amplify, and sequence ancient DNA (aDNA) from horse bone and teeth were optimized to recover DNA from a depositional environment of highly permeable acidic soil. DNA was successfully retrieved using 0.10g of bone powder from horse (Equus sp.) remains dating to 25 K years utilizing the methods optimized for this archaeological material. The genetic analyses were performed in a facility that is dedicated to ancient DNA research (Paleo-DNA Laboratory, Thunder Bay, ON, Canada) and has not been previously used to analyse modern or ancient horse DNA. Research was replicated to obtain reliable sequencing results for six samples from the Iberian Peninsula that were consistent with published sequences of Equus caballus. The archaeological sequence data obtained support hypotheses that promote the significance that the Iberian Peninsula has had to the multi-focal centres of origin for horse domestication and distribution of modern horse breeds. The data presented may provide evidence of the existence of an Iberian refugium for Equus during the last glacial period, 10 K years BP. Further molecular data analyses will enhance the ideas presented by this data and our understanding of horse domestication and phylogeny. The optimization of molecular techniques to successfully obtain DNA using minimally destructive, cosmetically sensitive techniques from archaeological remains endeavours to foster further cooperation between museums and researchers.  相似文献

Metacarpal radiogrammetry has received increasing interest in recent studies of archaeological human bone in facilitating the identification of age-related cortical bone loss and osteoporosis risk in the past. The technique is relatively simple and non-destructive, and existing comparative clinical data is an advantage in its application to archaeological samples. In order for metacarpal radiogrammetry to be useful in identifying individuals that could have been at risk of fracture in the past, cortical bone loss as measured at the second metacarpal is assumed to be reflective of bone loss occurring throughout the skeleton and across the metabolically different cortical and trabecular bone. Archaeological studies have frequently relied on literature reviews of clinical data to justify the technique as valid and useful. However, the present study has examined whether cortical bone loss measured in the second metacarpal is similar to bone loss measured at specific skeletal sites that are more frequently affected by osteoporosis-related fracture. Skeletons from two post-medieval collections from London, UK, Redcross Way and Farringdon Street (lower burial ground of St. Bride's Church) were studied. The results of the present study demonstrate that the second metacarpal is useful in identifying age-related bone loss at sites that are predilected to osteoporotic fracture, most notably the distal radius, which is prone to Colles' fractures. Metacarpal radiogrammetry also correlates well with bone loss in skeletal sites that are primarily composed of trabecular bone, including the fourth lumbar vertebra (a site of compression fractures) and the iliac crest (the location for clinical biopsies). The present study also confirms previous findings of significant differences between the pattern of cortical bone loss between the metacarpal and the femur. The poor relationship between these two areas is most likely attributable to mechanical loading.  相似文献

Abstract

The achievements, triumphs and disasters of leading northern semi-professional sportsmen were already being celebrated in broadsides at the start of the nineteenth century, and with the rise of when music hall entertainment became popular songs featuring such sportsmen became an important part of popular culture in Northumberland, Durham and Cumberland, the three Border counties. This article first shows what we know of the surviving songs and their performers, and then explores the ways in which such songs presented a picture of the nature and characteristics of northern sporting celebrity, in the context of a complex variety of local and regional sporting identities, especially in relation to other towns in the North and, more particularly, London. Sporting songs focused on individual success. They rejected the amateur ideals of athleticism, especially in terms of betting, and celebrated physicality, manliness and having a good time. Rowing was the most common focus of songs on Tyneside, wrestling in Cumberland, while horse racing, pedestrianism and boxing were also regularly covered.  相似文献

Parchments comprise one of the most common and valuable sources of archaeological and historical data. Previous studies have shown that parchment also preserves genetic data. These data could be valuable for population studies, to understand past animal husbandry, the development of breeds and varieties and to comment on the provenance of parchments. To improve our understanding of DNA contained in parchments, we analysed genetic data, including both mitochondrial and autosomal loci, from 18th to 19th century English parchments which stable isotope analysis had indicated were well-preserved. DNA results were unexpected. All but one of the parchments produced multiple sequences matching several different species. Ion beam analysis ruled out surface treatments of the parchments (including ink and animal glues) as the origin of these multiple sequences. Our results suggest that the DNA content of parchment is more complex than previous research has suggested and that multiple stages of parchment manufacture, treatment and storage are preserved in parchment DNA extracts.  相似文献
