This paper narrates the history of the conservation of the human brain, tracking techniques of brain archiving from the first experiments in the preservation of soft tissue in spirits of alcohol to the latest refinements in cryogenic technology. It traces the changing social and legal conditions that permitted the collection of post mortem human tissue, as well as the increasingly sophisticated technologies that allowed for the preservation and storage of this material. This preliminary survey of brain archiving uses examples of specific collections in order to discuss changes in the techniques, goals and achievements of neural tissue collecting from the mid-eighteenth to the late twentieth centuries.  相似文献

Teeth are often the preferred source material for isotopic and genetic assay involving ancient biomolecules. The assumption is that dental tissue preserves better due to its anatomically protected location, the enamel cap, and lower porosity compared to bone. However, this assumption has not been widely tested. Some similarities in diagenetic processes are to be expected due to similarities in structure and chemical composition of dentine and bone. This has led to the suggestion that bone can be used as an indicator of dental preservation, as a pre‐screening technique in the selection of suitable samples for biomolecular studies. Thus, direct testing of the correlation between bone and tooth preservation and diagenesis is needed. This paper reports the results of the histological characterization of diagenetic alterations within 25 human femur–tooth pairs, from a Medieval to modern (AD 1850) cemetery in Eindhoven, the Netherlands. The results showed that teeth do indeed preserve better overall, but not always, and that this was dependent on the main diagenetic factor(s) at the burial location. Furthermore, good correlations are found between the microstructural preservation of bone and teeth; similar processes of decay were observed within bone and teeth of the same individual. Overall, the study demonstrated that histological analysis of bone is useful for the identification of degradation processes that affect biomolecular preservation in skeletal material. Estimation of age‐at‐death is an important and challenging part of any investigation of human remains. Extensive research has been undertaken on this subject as demographic information contributes much to bioarchaeological and forensic work. Disarticulated, fragmentary and commingled human remains occur from a wide range of spatial and temporal contexts, and estimation of age‐at‐death can be particularly challenging in these collections. This study evaluated the impact of preservation on techniques that might be applied and their relative utility, using human remains from the site of the Smith's Knoll associated with the Battle of Stoney Creek, a War of 1812 collection from Ontario Canada with some supporting documentary evidence on age‐at‐death. Features assessed were the pubic symphysis, auricular surface and epiphyseal fusion in the innominate. An age‐at‐death estimate was produced for 16 out of 19 individuals considered; the highest minimum number of individuals (MNI) was 24 (right radius) from 2701 identifiable fragments. The pubic symphysis made no contribution to construction of a demographic profile, but as has been suggested previously epiphyseal fusion was useful in this respect. Results demonstrate that previous statements regarding preservation of the auricular surface and its utility in fragmented and poorly preserved collections need to be carefully evaluated. Although 129 fragments of innominate were recorded just one had an auricular surface and post auricular area that could be fully assessed. Transition Analysis was easier to apply than techniques developed previously, and findings from this study suggest that use of the forensic prior distribution could assist in assessment of battlefield assemblages. These results raise some valuable points that need to be considered in any future attempts to improve age‐at‐death estimates using the auricular surface, and important questions regarding expectations for estimation of age‐at‐death in disarticulated, fragmented and commingled collections of human bone. The search for the origins of syphilis has a long history in the medical and anthropological literatures. If we know more about the emergence of the pathogen that causes the disease in humans we will understand its evolution through time and space as well as shed light on its current state in living populations. Ancient DNA techniques used to isolate Treponema pallidum subsp. pallidum DNA from archaeological human specimens provide direct evidence of its existence in the past. However to date, only Kolman et al. (1999) have been successful in this endeavour, while other attempts have failed (e.g., Barnes and Thomas, 2006; Bouwman and Brown, 2005). Why has there been little success? This paper serves to compliment and add relevant information to Bouwman and Brown's and Barnes and Thomas' discussion concerning our inability to apply ancient DNA techniques to study venereal syphilis in past human populations.Our approach utilized 15 different human specimens from different geographies and different temporal periods: eight samples come from medically diagnosed individuals archived during the American Civil War period; six originate from the United Kingdom and predate 1492 with four of these samples having been previously analyzed by Bouwman and Brown and one sample comes from historic Canada. Human mitochondrial and amelogenin DNA, as well as several genes from the Treponema organism were analyzed revealing the relatively good preservation of human multi-copy and single copy DNA but not treponemal DNA. This study also incorporates a unique molecular experiment using rabbits infected with venereal syphilis to help illustrate that treponemal DNA disseminates to bone early during the first stages of infection but is not present in later stages of the disease using the techniques presented in this study.  相似文献

‘Historical’ DNA obtained from specimens preserved in natural history collections has proven useful for addressing a wide variety of questions, such as the spread of domesticated species or changes in genetic diversity. With the development of high‐throughput sequencing techniques, there is an increasing focus on acquiring genetic information encoded by single‐copy nuclear DNA from historical DNA extracts. The development of efficient techniques to determine the level of nuclear DNA preservation in candidate specimens is necessary to maximize the data obtained from these analyses. Although current evidence suggests that a sample's mitochondrial DNA preservation predicts its nuclear DNA preservation, we show that the relationships between mitochondrial and nuclear DNA preservation are complex.  相似文献

ABSTRACT

The ability to maintain human brain explants in tissue culture was a critical step in the use of these cells for the study of central nervous system disorders. Ross G. Harrison (1870–1959) was the first to successfully maintain frog medullary tissue in culture in 1907, but it took another 38 years before successful culture of human brain tissue was accomplished. One of the pioneers in this achievement was Mary Jane Hogue (1883–1962). Hogue was born into a Quaker family in 1883 in West Chester, Pennsylvania, and received her undergraduate degree from Goucher College in Baltimore, Maryland. Research with the developmental biologist Theodor Boveri (1862–1915) in Würzburg, Germany, resulted in her Ph.D. (1909). Hogue transitioned from studying protozoa to the culture of human brain tissue in the 1940s and 1950s, when she was one of the first to culture cells from human fetal, infant, and adult brain explants. We review Hogue’s pioneering contributions to the study of human brain cells in culture, her putative identification of progenitor neuroblast and/or glioblast cells, and her use of the cultures to study the cytopathogenic effects of poliovirus. We also put Hogue’s work in perspective by discussing how other women pioneers in tissue culture influenced Hogue and her research.  相似文献

Ancient bones in a good preservation state, ascertained by microscopic techniques, conserve extracellular matrix proteins over thousands of years. With new techniques, intact extracellular matrix proteins from ancient bones and teeth are extracted and separated by one‐dimensional and two‐dimensional electrophoresis. Proteins were identified in Western blots by special antibodies against different human extracellular matrix (ECM) molecules of bone. We have confirmed different types of ECM human bone molecules such as osteonectin, osteopontin, and alkaline phosphatase with specific antibodies in human bone samples from different age groups. Additionally, we selected bone samples from different cultural time periods, such as the Middle Ages, the Bronze Age and the Late Pre‐Pottery Neolithic Phase (PPNB), and teeth from individuals from the Early Middle Ages and from the Late PPNB. The survival of intact extracellular matrix proteins in ancient bones and teeth dating from recent times to the Late PPNB, and reliable techniques to identify these proteins, present a great challenge to further research. A Match Set with PD‐Quest 7.2 shows that only 16% of protein spots in the teeth are also found in the bone of the same individual. In combination with the results of macro‐ and microscopic investigation, biochemical techniques will help us in obtaining a better understanding of bone and teeth in health and disease. Using sensitive techniques of molecular biology, we have been able to demonstrate the presence of genomic material of human origin in samples of mummified human tissue and bone from selected archaeological sites in Greenland. In the ground, bone undergoes chemical and physical changes which affect its preservation. This fact has important implications for dating and other analytical procedures involving bone, as well as faunal analysis where differential preservation of bones of different species may affect conclusions regarding the relative significance of an animal to the economy of a given society. The diagenic processes in bone range from minor changes in the bone protein to complete structural and chemical breakdown.Using fresh cow bone, we conducted laboratory experiments which simulate the effect of temperature and bone size on the rate and nature of bone disintegration in archaeological sites. Temperature influences the rate of chemical change, and bone size and density affect the accessibility of the molecular constituents of bone to extrinsic chemical reactions. These findings clarify the importance of two well-known concepts in bone taphonomy. (1) The rate of chemical breakdown in bone tissues is related to the proximity of a given unit of tissue to the bone surface. This means that, in archaeological bone samples, tissue near the surface may be different chemically from tissue away from the surface and great care is necessary in choosing and preparing bone samples for analytical procedures. (2) In general, small bones are not as well preserved as large bones, therefore small animals are likely to be underrepresented in faunal assemblages.  相似文献

Materials and methods for the recovery and analysis of microscopic plant remains in underwater archaeology. This article explores formation of underwater sites, preservation and potential contamination of botanical remains, sampling techniques, benefits of sampling and searching for microscopic remains including pollen, phytoliths, cystoliths, starch grains, epidermal tissue, and cellulose fibre, and conservation and archiving of archaeobotanical samples. Materials such as ship's caulking, surrounding sediment matrices, bilge sediments, and organic remains found inside various types of cargo containers can contain microscopic plant remains that become botanical fingerprints used to identify cargoes, ship's foods, onshore vegetation, location of a ship's home port, and plants used to make rope, basketry, and matting.  相似文献

Henry Herbert Donaldson (1857–1938) was a leader in neurological research in the United States for several decades, beginning about 1890. A detailed account of three of his earliest publications shows the neuroanatomical procedures involved in the study of the relation of brain and intelligence during the late-nineteenth-century in America. Two of the articles, published in September 1890 and December 1891, were titled, “Anatomical Observations on the Brain and Several Sense-Organs of the Blind Deaf-Mute, Laura Dewy Bridgman (1829–1889)”; the third, published in August 1892, used the information from the first two to delimit the extent of the visual processing area of the human cortex. Donaldson’s procedures included brain cuttings and measures of macroscopic brain structures, histology of cellular structures, attempts to relate macroscopic brain structures with brain functions, data corrections, estimations, comparisons, and statistics. These procedures provide a view of the relative thoroughness, accuracy, and comparability of the various neuroanatomical techniques in use at that time and of Donaldson’s implementation of the techniques. Donaldson’s brain cutting techniques were much more comparable than his measurement techniques. The latter could be quite precise, but they were fraught with lack of standardized procedures that made corrections and estimations necessary when making data comparisons across studies. Donaldson emphasized these incompatibilities, implying a need for standardization. Statistical procedures were the least thorough and effective. His, and the field’s, total complement of statistical techniques consisted of mean and range, which severely limited his ability to make complicated assessments. This limitation was not necessarily supplemented by stringent control group comparisons.  相似文献

This article presents a comprehensive overview of the techniques available for the seismic retrofitting of adobe buildings. It analyzes these techniques’ viability from a structural engineering standpoint and evaluates their suitability in the context of historic preservation. Analysis was based on an exhaustive review of studies addressing retrofitting techniques of earthen structures, as well as consultation with field experts. The guiding principles of conservation relating to the structural reinforcement of monuments were taken into account. Traditional techniques that enhance the stability of the building, such as wooden ring beams, wooden ties interconnecting parallel walls, corner keys, and the addition of buttresses were found to be effective solutions since they employ compatible and low-cost materials such as earth and wood. When these techniques prove insufficient, minimally invasive measures such as introducing a plywood diaphragm, horizontal steel rods, a geomesh covered with mud rendering or a strapping system can be appropriate.  相似文献

A baseline survey in 2007 characterized the physical and chemical conditions for preservation within waterlogged remains at Nantwich. Installation of eighteen dipwells has allowed a five-year monitoring programme to be conducted from 2011. Two add-on projects supplemented this monitoring programme by examining the different methods for redox measurement, and for soil moisture measurement using Time Domain Reflectometry (TDR).

Initial results show two main zones of preservation, with reducing conditions attributed to location of the deposits in the floodplain, and a second zone uphill with more variable conditions for preservation. This study shows that sediment-coring programmes combined with dipwell installations can provide useful data to assess and monitor in situ preservation conditions, which can help to formulate management strategies for conservation of waterlogged archaeological deposits. This paper focuses on the efficacy of the methodology, referencing in particular comparative monitoring techniques and lists a series of recommendations for future studies.  相似文献

The rich mass deposition of Iron Age human bone material from the Danish site, Alken Enge, is extraordinary not only from an archaeological perspective but also from a preservation point of view. The main find is situated in a waterlogged, anoxic environment which provides excellent preservation conditions and therefore enables in situ preservation of the human bones. However, major differences in local environmental conditions challenge an in situ preservation of the entire site area as parts of the bone material presently deteriorate. In this paper, a multi-proxy environmental monitoring approach is used to document threats and to suggest the best preservation solution for the archaeological finds, whether in or ex situ.  相似文献

The localization or representation of mental abilities in the brain have always been considered as key questions for understanding the organization of the human nervous system. Particularly with the advent of modern electrophysiological and imaging techniques that provide maps of electromagnetic fields and metabolic processes on the living central nervous system, the representation theory is experiencing a scientific renaissance in neurology, but is only one theory, however, in the succession of a long philosophical tradition dealing with the possible identification of mental phenomena and brain processes. This dichotomy was formulated at the latest in the Cartesian dualism of res cogitans and res extensa of the mind-body problem. Nowadays philosophical discussion, on the contrary, is dominated by monistic concepts that attempt to explain the mental realm on an organic foundation in order not to succumb to the problem of a psychophysical dualism. Of these, the identity theory offers a philosophically plausible concept postulating that the identity of brain conditions and mental phenomena is based on organic foundations. In this theory, the efforts of brain research converge on the representations of mental phenomena in the human nervous system. In a comprehensive approach, both concepts could complement each other.  相似文献

Archaeological work in advance of construction at a site on the edge of York, UK, yielded human remains of prehistoric to Romano-British date. Amongst these was a mandible and cranium, the intra-cranial space of which contained shrunken but macroscopically recognizable remains of a brain. Although the distinctive surface morphology of the organ is preserved, little recognizable brain histology survives. Though rare, the survival of brain tissue in otherwise skeletalised human remains from wet burial environments is not unique. A survey of the literature shows that similar brain masses have been previously reported in diverse circumstances. We argue for a greater awareness of these brain masses and for more attention to be paid to their detection and identification in order to improve the reporting rate and to allow a more comprehensive study of this rare archaeological survival.  相似文献

赭石的使用是现代人行为的标志之一,是除石制品之外更能反映现代人认知能力等深层次精神世界的载体,但是国内对该类物质的研究还比较少。本文主要参照西方学者的研究成果,讨论中国遗址中赭石的功能,认为赭石在旧石器时代的中国地区主要被用作颜料、兽皮防腐和药物,同时它还被用于原始宗教活动。与旧大陆西侧遗址中的赭石相比,除了相似的使用功能,西方的古人类利用赭石的时间更早,所以本研究推测中国地区古人类对赭石功能的认识可能源于西方并在现代人迁徙的过程中流入中国地区。这些证据指示着中国地区的现代人由北向南的迁徙路线。  相似文献

Although all of the Korean medieval mummies found up to the present time have shown relatively well‐preserved brains, relevant biomedical information has been difficult to acquire owing to the obstacles to obtaining permission to conduct invasive investigations. However, recently we were able to secure permission to investigate, through various forms of examination including dissection, a well‐preserved brain found in a lime‐soil mixture barrier (LSMB) tomb in Yongin, Korea, because the remains found within the tomb were mostly skeletal. As in the cases of mummified brains from tombs found in other countries, the current Yongin case showed well‐preserved grey and white matter within well demarcated brain lobes, on which sulci and gyri could be identified. On histological examination, we found that the remaining brain tissue was composed mainly of lipids, which seems to correspond to the preserved myelin sheath. The present paper is the first report on the general preservation status of a mummified brain found in Korea. Moreover, because mummified brains are found quite frequently in LSMB medieval tombs, this study could provide a good basis upon which further such palaeo‐neuropathological studies can be progressed. Abstract

Underwater Cultural Heritage (UCH) as an outstanding division of the cultural heritage of humanity appears to be crucial and complicated when more general issues regarding preservation and conservation are raised. The essence of in situ preservation should be equally discussable for any kind of archaeological remains; on land or underwater.

There is a long history of different methods and concepts of intervention in a variety of sub-aquatic archaeological sites; from shipwrecks to submerged settlements. This paper will present an introduction to different techniques and theories of preservation and conservation of underwater cultural and archaeological sites since this kind of heritage has scientifically been explored and studied. A range of different preservation methodologies, from total or partial transference inland, to preservation underwater, will be compared; the advantages and disadvantages of each option will be highlighted. Different examples of international best practices will be illustrated. Different types of in situ conservation/protection will be explained and categorized. Furthermore, there will be a focus on the UNESCO Convention of 2001 on Conservation and Preservation of UCH, where the in situ conservation option has been recommended.

Moreover, the technical issue for preservation of UCH sites, either in situ or after displacement, will be explained. The implication of relocation for different sorts of sites and materials will be argued; for example, cases where some sites, such as shipwrecks, would more easily be displaced compared with submerged settlements, villages, or ports.

Finally, by stressing that the state of ‘being underwater’ makes many sites qualified to be regarded as UCH, the in situ preservation approach will prevail that this state is maintained.  相似文献
